Professional biography: Dr. Soontaree Petchdee is an Associate Professor in the Faculty of Veterinary Medicine at Kasetsart University (Kamphaeng Saen campus). Her research focuses on veterinary cardiology, cardioprotection, diagnostic echocardiography, and translational cardiovascular models in small and large animals. She holds a PhD from the University of Glasgow and leads clinical and experimental projects, including work on mitral valve repair techniques in dogs and cardioprotective natural-product research.

Lecture Summary

Saponins obtained from the sea cucumber Holothuria leucospilota have been widely recognized for their metabolic and cytoprotective properties; however, their influence on cardiovascular structure and function has not been fully clarified. This study investigated the cardioprotective effects of saponin supplement in a high-fat diet rabbit model with renovascular hypertension. Sixteen New Zealand White rabbits were divided into four experimental groups: a normal diet group, a high-fat diet group with partial renal artery ligation, and two treatment groups receiving the same high-fat diet and ligation combined with either low-dose or high-dose saponin supplementation. Hypertension was established through partial constriction of the left renal artery.

Physiological and metabolic assessments, including body weight, blood biochemical profiles, arterial pressure, and cardiac structure and function, were conducted at baseline and during the experimental period. Rabbits receiving saponin supplementation demonstrated an improvement in metabolic indices, with significant reductions in circulating glucose, triglycerides, and cholesterol levels compared with untreated hypertensive animals. In addition, mean arterial pressure was substantially reduced in both saponin-treated groups. Cardiac imaging revealed a clear attenuation of left ventricular wall thickening, indicating suppression of hypertensive cardiac remodeling.

These findings provide evidence that saponins derived from Holothuria leucospilota exert protective effects on the heart under conditions of diet-induced metabolic stress and elevated blood pressure. The combined improvement in metabolic status, blood pressure regulation, and cardiac structure suggests that this marine-derived compound may represent a promising natural adjunct strategy for reducing cardiovascular risk associated with obesity and hypertension.
